Ipamorelin dosage in studies: doses, timing and reconstitution

The human doses reported were 0.03 mg/kg by intravenous infusion twice daily for up to 7 days (Beck et al. 2014) and single 15-minute infusions of 4.21–140.45 nmol/kg (Gobburu et al. 1999). Rat studies used intravenous doses of 0.01–1 mg/kg and subcutaneous doses of 18–450 micrograms/day or 100 micrograms/kg three times daily.

What dose of ipamorelin have studies used? In people, two doses are reported. Beck et al. (2014) gave 0.03 mg/kg by intravenous infusion twice daily, from the first day after bowel surgery until day 7 or discharge. Gobburu et al. (1999) gave single 15-minute intravenous infusions at five levels, from 4.21 to 140.45 nmol/kg, to eight healthy men at each level. Both were intravenous; neither study used subcutaneous dosing.

The human pharmacokinetic study explains why frequency matters in study design. Ipamorelin had a terminal half-life of about 2 hours, and each infusion produced a single growth hormone pulse that returned to negligible levels. Longer rat studies therefore split the daily amount into several injections: three times daily in Johansen et al. (1999) and Andersen et al. (2001), or continuous infusion by osmotic minipump in Svensson et al. (2000).

What doses were used in rat studies? For postoperative ileus, Venkova et al. (2009) gave 0.01–1 mg/kg intravenously, either once or four times a day for 2 days, and Greenwood-Van Meerveld et al. (2012) gave 0.014–0.14 µmol/kg intravenously. For bone and growth, Johansen et al. gave 18, 90 or 450 micrograms/day subcutaneously for 15 days, Svensson et al. gave 0.5 mg/kg per day for 12 weeks, and Andersen et al. gave 100 micrograms/kg three times daily for 3 months.

The studies report doses in different units: nmol/kg and µmol/kg in some, mg/kg or micrograms per day in others. Compare them only after converting to a common unit, and keep in mind that doses in rats do not transfer directly to other species.

What about CJC-1295 and ipamorelin together? None of the studies in our ipamorelin records tested the pair, so there is no published combined dose to report. Tasman supplies ipamorelin as a 10 mg vial and, separately, a premixed 10 mg research vial of CJC-1295 (no DAC) 5 mg + ipamorelin 5 mg. For laboratory reconstitution the site's calculator uses 1 mL of bacteriostatic water per 5 mg as a neutral maths default, so 2 mL for a 10 mg vial. This is arithmetic only, not a dose.

What these doses don't tell you

There is no human dose-finding study for repeated subcutaneous use in these records; both human studies used intravenous infusion. No study here tested ipamorelin combined with CJC-1295, so no combined dose has been published in these records.
