What is Ipamorelin?
Ipamorelin is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) that acts on the growth hormone secretagogue receptor, also called the ghrelin receptor. It came out of a 1990s chemistry programme on compounds derived from GHRP-1 and was described in 1998 as the first selective growth hormone secretagogue. In rats and pigs it released growth hormone about as strongly as GHRP-6, but unlike GHRP-6 and GHRP-2 it did not raise ACTH or cortisol, even at doses more than 200 times the dose needed for half its growth hormone effect. Because the ghrelin receptor also affects gut motility, it was later tested in rats and in one human phase 2 trial for postoperative ileus. Most of the published work is in rodents, and the human data are limited to short studies.

What these studies don't show: The only controlled human trial here is a small phase 2 study in post-surgical patients, and it found no significant benefit over placebo on its efficacy endpoints. The other human study measured pharmacokinetics and growth hormone release after a single infusion in healthy men. The bone, muscle and growth results come from rats, and there are no long-term human data on repeated use.
