5-Amino-1MQ dosage in studies: doses, timing and reconstitution

Only one abstract on file gives a dose: 5 or 10 mg/kg of an NNMT inhibitor in aged mice for 1 or 3 weeks after muscle injury, and that abstract does not name the compound. The obese-mouse study of 5A1MQ used once-daily dosing for 28 days without stating the amount, and the HeLa cell study used 0.1–500 µM. These are reported study doses only.

What doses of 5-Amino-1MQ have studies used? The honest answer is that most abstracts on file do not say. The one stated animal dose is 5 or 10 mg/kg in Neelakantan et al. 2019, given to 24-month-old mice for 1 week (both doses) or 3 weeks (10 mg/kg only) after a muscle injury. The abstract calls the compound an NNMT inhibitor developed by the same group and does not name it, so that figure cannot be tied to 5-Amino-1MQ with certainty.

Babula et al. 2024 gave 5A1MQ once daily for 28 days to diet-induced obese mice and reported dose-dependent effects, which means more than one dose was tested, but the abstract does not give the amounts or the route. Their pharmacokinetic arm dosed mice by intravenous, oral and subcutaneous routes, and subcutaneous dosing distributed the compound to fat, muscle and liver.

In rats, Awosemo et al. 2021 measured plasma levels after intravenous and oral doses and reported a mean oral maximum concentration of 2252 ng/mL, again without stating the dose in the abstract. In HeLa cells, Akar et al. 2021 tested 0.1 to 500 µM. A cell-culture concentration does not translate into an animal or human dose.
